Serotonin neurons in the median raphe nucleus bidirectionally regulate somatic signs of nicotine withdrawal in mice

چکیده
In chronic smokers, nicotine withdrawal symptoms during tobacco cessation can lead to smoking relapse. rodent models, exposure elicited physical dependence, whereas acute antagonism of nicotinic acetylcholine receptors (nAChRs) immediately precipitated symptoms. Although the central serotonergic system plays an important role in withdrawal, exact raphe nuclei regulating these remain unknown. We used transgenic mice expressing archaerhodopsinTP009 or channelrhodopsin-2[C128S] exclusively neurons selectively manipulate each nucleus. Nicotine were by injection mecamylamine, a nonspecific nAChR antagonist, following consumption. Somatic signs as measures Acute mecamylamine administration significantly increased ptosis occurrence nicotine-drinking compared with that control-drinking mice. Optogenetic inhibition median nucleus (MRN), but not those dorsal (DRN), mimicked observed mecamylamine-precipitated even nicotine-naïve injection. activation MRN nearly abolished The MRN, DRN, are necessary for somatic signs, symptom, and may act potential therapeutic strategy preventing manifestations withdrawal.

Serotonin neurons in the dorsal raphe nucleus encode reward signals.
The dorsal raphe nucleus (DRN) is involved in organizing reward-related behaviours; however, it remains unclear how genetically defined neurons in the DRN of a freely behaving animal respond to various natural rewards. Here we addressed this question using fibre photometry and single-unit recording from serotonin (5-HT) neurons and GABA neurons in the DRN of behaving mice. Rewards including suc...
متن کاملEngrailed is required in maturing serotonin neurons to regulate the cytoarchitecture and survival of the dorsal raphe nucleus.
Analysis of constitutive Engrailed (En) null mice previously implicated the two En homeobox paralogs in the development of serotonin (5-HT) neurons. An unresolved question is whether En plays intrinsic roles in these neurons. Here, we show that En1 and En2 are expressed in maturing 5-HT neurons that will form the dorsal raphe nucleus (DRN) and part of the median raphe nucleus. Although En1 expr...
متن کاملAcute selective serotonin reuptake inhibitors regulate the dorsal raphe nucleus causing amplification of terminal serotonin release
Selective serotonin reuptake inhibitors (SSRIs) were designed to treat depression by increasing serotonin levels throughout the brain via inhibition of clearance from the extracellular space. Although increases in serotonin levels are observed after acute SSRI exposure, 3-6 weeks of continuous use is required for relief from the symptoms of depression. Thus, it is now believed that plasticity i...
متن کاملProjections of the median raphe nucleus in the rat.
No previous report in any species has examined comprehensively the projections of the median raphe (MR) nucleus with modern tracing techniques. The present report represents an in depth analysis of the projections of MR by use of the anterograde anatomical tracer Phaseolus vulgaris-leucoagglutinin. MR fibers descend along the midline within the brainstem and mainly ascend within the medial fore...
متن کاملHypocretins (orexins) regulate serotonin neurons in the dorsal raphe nucleus by excitatory direct and inhibitory indirect actions.
The hypocretins (hcrt1 and hcrt2) are expressed by a discrete population of hypothalamic neurons projecting to many regions of the CNS, including the dorsal raphe nucleus (DRN), where serotonin (5-HT) neurons are concentrated. In this study, we investigated responses to hcrts in 216 physiologically identified 5-HT and non-5-HT neurons of the DRN using intracellular and whole-cell recording in r...
